Design Notes
The part has multi cloning sites containing NdeI, HindIII, BglII, ClaI, and BamHI.
You can construct complete sequences and negative control easily using these restriction sites. For example, you cut BamHI site, you can remove backward SAR.
You can amplify this fragment following two primers.
Forward: GGGGAATTCGCGGCCG(starting from EcoRI and NotI sites)
Reverse: CCCACTAGTTATAAACGCAGAAAGGCCC(starting from SpeI site)
